Title: Cancer Genome Analyses to Personalize Treatment: From Microarray to Next-generation sequencing data
Abstract:
Recently developed high-throughput biotechnologies allow mapping of the complete genome sequence, and molecular and cellular features of human cancer. Finding associations between human cancer and various ‘omics data including gene expression, DNA copy numbers, and next-generation sequencing data can shed light on the molecular and cellular mechanisms underlying human cancer, but calls for new computational approaches for its analysis.
To address these challenges, we have developed and applied computational approaches to integrate various types of genomic data with prior knowledge (e.g., pathway, protein-protein interaction network and etc.) for biomarker discovery, cancer outcome prediction and patient stratification. In this talk, I will present our recent efforts (1) to integrate genomic data (e.g., gene expression, copy number, and mutation) with pathway to identify a) patient subgroups having different survival outcomes and b) pathways disrupted by somatic mutations in human cancer, (2) to estimate heterogeneity of cell populations linked to therapy resistance next-generation sequencing data. These methods and analyses can provide a systematic understanding of the mechanisms of cancer development and progression, and thus suggest the potential for a guide to targeted therapy in cancer patients. In addition, I will discuss lessons learned and new challenges in cancer genomics.
