Title: Epigenetic Landscapes at Gene Regulatory Regions

 

 

 

Abstract:

The eukaryotic epigenetic landscapes, composed of many epigenetic factors including histone modifications, DNA methylations and histone variants, control the function of cells by regulating gene activity. At regulatory regions, the combination of epigenetic factors represents the activity of the genome in association with transcription factors and their co-factors. However, our understanding about the epigenetic languages is still limited.

We comprehensively exploited the epigenetic landscapes drawn by histone variants, histone modification and DNA methylation using chromatin immunoprecipitation followed by massive sequencing (ChIP-seq) data. Using a systematic way, we found various languages (histone codes) cells use for gene regulation. Especially, we investigated the shape of histone variants and modification at gene regulatory regions. We found various regulatory rules represented by the shape and the combination of epigenomic data. Furthermore, we investigated the landscape of 5-hdroxymethylcytosine (5hmC) in association with histone modification. The computational approach as well as experimental validation revealed the role of 5hmC at gene regulatory regions.
