Title: Sequencing the Cancer Genome

 

Rapid advances in high-throughput sequencing technologies have made it possible to generate high-coverage whole-genome sequencing (WGS) data for a patient at an affordable cost. I will describe our analyses of structural variations in cancer genomes using WGS data from ~50 cancer patients (~150 billion short reads). In particular, we have detected a large number of retrotransposition events in selected tumor types, reconstructed complex structural variations from clusters of paired-end reads, and classified the mutational mechanisms based on sequence homology at the breakpoints. I will also discuss the challenges associated with large-scale data management and analysis

 

 

 

Bio : Dr. Peter Park is an associate professor at Harvard Medical School, where he leads a computational genomics group. Originally trained in applied mathematics at Harvard and Caltech, he was introduced to molecular biology and genetics during his postdoctoral studies in biostatistics. His laboratory specializes in understanding chromatin structure and its role in gene regulation as well as in characterizing the cancer genome, based on high-throughput sequencing data. His group (14 postdoctoral fellows, 3 doctoral students, and 2 programmers) is also involved in several consortium projects including The Cancer Genome Atlas (TCGA) and the Encyclopedia of DNA Elements (ENCODE). He is a co-director of the Bioinformatics and Integrative Genomics (BIG) Ph.D. program at Harvard and a member of the Dana-Farber Cancer Center and the Harvard Stem Cell Institute.
